Mechanism of action
Nabumetone selectively inhibits prostaglandin synthetase-2(PGHS-2). This isoenzyme is responsible for prostaglandin synthesis and thus inflammation in the tissue of such areas as diarthroidal joints. It is metabolised into 6-methoxy-2 naphthyl-acetic acid that is a potent inhibitor of PGHS-2. It has no inhibitory effect on PHGS-1 which is responsible for prostaglandin synthesis in gastric mucosa, thereby minimizing the risk of problems like ulcers and hypertension.
